Senator Stockwell (John Carter), monitoring the television launch of a Star-Flight Twelve, Saturn V. Rocket, is joined by Victor Connelly (Anthony Eisley) head of the CIA, as the TV reception is interrupted by a distorted vision and an anonymous voice asking the Senator if "he is watching well?" and immediately cautioning him that "next time perhaps you'll listen!"
Video is immediately resumed, only to have the rocket explode in a tremendous, devastating malfunction. The two grim spectators immediately launch an investigation, calling on Sabrina Kincaid (Francine York) leader of a shapely and deadly coterie of female lovelies known as "The Doll Squad" to head the inquiry. Sabrina is intrigued by the voice of the unknown intruder, and except for the fact that future negotiations were to be continued by means of carrier pigeons, there are no other clues.
Sabrina enlists the aid of Carol Pierce (Carol Terry) and Cherisse (Brett Zeller) a karate operator and pathologist, respectively, but both girls are killed by Munson (Herb Robbins) and a trio of killers. Sabrina barely escapes with her life after inflicting a horrendous burn on Munson by means of a trick cigarette lighter.
Sabrina's investigations ultimately reveal that Nancy Malone (Lisa Garret) secretary to the Senator has served as intermediary for Eamon O'Reilly (Michael Ansara) former CIA agent, previously reported dead, but who apparently is now heading this overt, treacherous action against the government.
Nancy's suicide apparently confirms this knowledge, and Sabrina now reactivates "The Doll Squad," a band of female specialists, each skilled in a particular field, and dedicated to the overthrow of America's enemies.
They are Sharon O'Conner (Leigh Christian), Cat (Sherri Vernon), Elizabeth White (Judy McConnell), and Lavelle Sumara (Tura Satana).
Sabrina has learned that Eamon O'Reilly is headquartered at a foreign island and now with her band she sets off in pursuit.
Meanwhile, on the island, Joseph (William Bagdad) who works for O'Reilly, contacts a Kim Luval (Jean London) an uninitiated member of "The Doll Squad" who has been waiting for Sabrina's arrival, posing as a cashier at a local carnival, dupes and captures her, replacing her with Maria (Lisa Todd) complete with replica mask and assuming the identity of Kim.
This ruse quickly fails when Maria is unable to answer Sabrina correctly. Sabrina overpowers Maria and drives off with her, leaving a frustrated Joseph to contact O'Reilly.
At their headquarters, the girls pinpoint O'Reilly's location from Maria, but in endeavoring to replace Maria for Kim, it is Maria who suffers a mistaken death in place of Kim. "The Doll Squad" sets out to track down O'Reilly, with Cat and Elizabeth pursuing an overland route by jeep and Sabrina and her squad via boat.
After Sabrina and her group debark the boat and head for their assignment, Rafael (Rafael Campos) secretly working for and in the clutches of O'Reilly due to a drug addiction, misinforms Captain Curran (Curt Mason) into returning later than previously arranged for, then sets out after the girls and actually tricks them into capture.
Cat and Elizabeth reach the outskirts of O'Reilly's desert fortress, overpower and destroy the guards with a self-destructive alcoholic beverage and press on.
At a meeting with all foreign representatives, Eamon O'Reilly now reveals his master plan; subjugation and overthrow of all foreign governments by means of artificial bubonic plague. All are to be inoculated against the disease, with twins, Dr. Cahaymen and Dr. Cahaymen (Gustave and Bertil Unger) in charge of this phase of operations.
Cat and Elizabeth reach their objective, just as Sabrina is taken by Rafael to meet for the first time with the ambitious O'Reilly. Sabrina quickly eliminates and kills the drug-crazed Rafael, then joins with Cat and Elizabeth in releasing the others. However, Kim is killed in this rescue.
Now Sabrina is free to follow through on her appointment with O'Reilly, as the rest of "The Doll Squad" continue on their assigned tasks; elimination of the balance of the armed guards and the implanting of dynamite charges to destroy the fortress and any and all agents. But Sabrina must first eliminate an angered Munson, who suddenly confronts her prior to her appointment.
In the meeting with O'Reilly, Sabrina and he discuss their former love affair, with each actually endeavoring to kill the other. Sabrina finally wins this battle of wits, by means of a mace ring and a sword.
Dr. Cahaymen is eliminated as he attempts to escape via airplane. As "The Doll Squad" resume their journey to safety, the entire desert fortress is eliminated in a tremendous explosion. Only Dr. Cahaymen and a few guards now stand in their way, but these are eliminated by means of a bazooka.
A puzzled Captain Curran marvels at the insouciance, affability and carefree attitude of the somewhat bedraggled crew who stumble back aboard his boat. Sabrina reports her success back to Senator Stockwell and Victor Connelly, with the full knowledge that further adventures still await her and "The Doll Squad."
